Penfluridol-antipsychotic drug and its pharmacology

Introduction
Penfluridol is a highly potent, first generation diphenylbutylpiperidine antipsychotic. It was discovered at Janssen Pharmaceutica in 1968.

Therapeutic Uses
-Analgesic
-Anti Convulsant
-Anti Depressant
-Anti Migraine
-Neuropathic pain
-Hypnotics


Side effects

-Blood dyscrasias
-Dry mouth
-Blurring vision
-Retention of urine
-Granular deposits in cornea
-Weight gain

Mechanism of action


Penfluridol blocks the postsynaptic dopamine receptor in the mesolimbic dopaminergic system and inhibits the release of hypothalamic and hypophyseal hormones.


Drug-drug interaction
1-Penfluridol+MAOIs= Orthostatic hypotension
2-Penfluridol+Metoclopramide= May increase risk of extrapyrimidal symptom
3-Penfluridol+Amphetamine= May increase psychosis
4-Penfluridol+Levodopa= May inhibit antiparkinsonian effect
