LANSOPRAZOLE-
MACHENISM OF ACTION
▪ Lansoprazole is a proton pump inhibitor.
▪ Lansoprazole is a prodrug, after administraion it diffuses into the parietal cell of the stomach and accumulates in the secretory canaliculi.
▪ In the acidic medium lansoprazole is converted to sulfenamide(active form).
▪ This sulfenamide covalently bind with sulfhdryl(SH) group in the proton pump(H+K+ATPase) and inhibit the exchange of extracellular K+ for intracellular H+ ion.
▪ Lansoprazole irrevesibly inhibits proton pumps activity and decreases gastric acid secration.
